Sterile Device Packaging: What to Check Beyond the Shipping Box
This practical question connects the final packaging composition and distribution conditions with post-transportation verification items.
It examines the application scope of existing validation evidence through a hypothetical scenario involving a change in cushioning material.
Distinguishing Between the Box Exterior and Internal Packaging
It is easy to feel assured if there are no dents in the shipping box.
But can we conclude that the sterile barrier inside has been confirmed just by inspecting the exterior of the outer box? This article presents review questions for teams developing final sterile medical devices or changing packaging compositions.
It does not replace product-specific test plans or shipping judgment procedures.
According to the FDA’s public training materials for 2024, maintaining the packaging in a sterile condition after sterilization is crucial.
This article will use that explanation as a starting point and propose self-questions that connect the actual distribution process with development data.
FDA Original Text, Packaging Integrity Explanation, PDF pages 6-8
First, figure out what the term 'packaging' means in the context your team is using.
If we call the transportation box, cushioning, product securing structure, and sterile barrier by one word, the scope of change or the evaluation target can be misunderstood differently.
It is useful to open the actual product's packaging drawings and composition charts and indicate which parts are being discussed.
Distinguishing Between Two Evaluation Questions
The FDA describes packaging performance evaluation as questions regarding the effects of manufacturing, sterilization processes, and distribution and storage environments, whereas packaging stability evaluation concerns whether barriers are maintained over the shelf life.
Don’t stop at just distinguishing the names of these two perspectives; examine what questions the results you have actually answer.
MedQraft’s practical suggestion: Write briefly beside the report list ‘the scope described by this data.’
For example, distinguish whether the data concerns the current packaging composition or refers to data from another model.
It is better to separately record the fact that data exists and the judgment that it can be applied to the current product/composition.
Information to Organize Before Test Requests
The following table is a meeting review framework created by MedQraft.
It is not a comprehensive requirement list of legal forms or specific specifications.
| Review Item | Questions to Verify Together |
|---|---|
| Product | What are the model, shape, weight, and arrangement within the packaging? |
| Composition | What is the shipping box, cushioning, securing structure, and sterile barrier that will actually be shipped? |
| Path | How is the product moved and stored along each segment? |
| Representativity | What product range and packaging composition does the selected test sample represent? |
| Verification Results | What methods and criteria will be used to assess the results after testing? |
During the process of filling out the table, leave parts you do not know as assumptions.
For example, if the delivery path after the distributor is not organized, it’s better to determine who to verify and what materials to check instead of closing it with ‘no problem.’
Before promising to reproduce every segment, clarify the reviewed scope and remaining uncertainties.
Hypothetical Example: If the Pouch Stays the Same but Only the Cushioning Material Changes
The following is a hypothetical situation for explanation, not a real customer case.
A manufacturer wants to change the cushioning material of the shipping box while using the same sterile pouches.
The procurement officer understands it as a material change, while the development officer understands it as a change in product securing methods.
In this case, instead of concluding ‘retesting needed’ or ‘no testing required’ right away, we can examine the differences in the changed aspects.
Look into whether the product has room to move inside the box, the locations where protrusions touch, and whether the direction in which the packaging worker inserts the products changes.
This is an example of questions we can consider regarding the change, and it does not imply that actual risks or defects have been identified.
If existing data is judged to apply to the new composition, provide reasons for that.
If additional evaluation is necessary, link it to the evaluation aimed at resolving any uncertainties.
Avoid ordering a generic test package simply because there's a change, or concluding the review just because the pouch material remains the same.
What to Compare Before and After Transportation
MedQraft’s practical suggestion: Don’t leave the last column of the test request form as ‘receipt of results’; determine who will review the findings and what decisions will be based upon them.
The items below are self-written examples.
| Stage | Records to Leave |
|---|---|
| Before Starting | Identification of test items, packaging configuration, readiness status, and applied documents |
| Conditions Setup | Selected distribution conditions and reasons for choosing those conditions |
| Results Verification | Observational and measured results, applied criteria, any deviations |
| Conclusion | The scope described by the evidence and unresolved questions |
| Follow-Up Actions | Responsible individuals for additional verifications, required materials, review timing |
Exterior photos can be challenging to compare if the position or direction changes.
When creating internal team records, it is advisable to include explanations of what the photos aim to show.
Do not interpret photos as automatically replacing other evaluations.
Don’t Treat Test Method Names as Answers
The FDA training materials explain that while various packaging evaluation methods are mentioned as examples, one cannot only use the listed tests.
Thus, avoid the approach of copying test method names from public presentations without product review.
FDA Public Explanation, PDF page 7
Actual methods, sample sizes, application order, and passing criteria should be aligned with the product, packaging materials, intended distribution conditions, and market requirements.
This article does not specify particular temperature, vibration, drop conditions, or sample sizes.
It also does not express reviewing specific provisions without possessing a standard specialized text.
When coordinating with test institutions, prepare materials that explain why this method is suitable for this question rather than simply asking which test is famous.
Three Points to Address in the Next Meeting
First, identify the final packaging configurations before and after the change using the same criteria.
Second, distinguish the distribution and storage conditions described by existing evidence and those to be verified this time.
Third, determine additional questions to confirm, and assign individuals responsible for conclusion-making.
This is MedQraft’s business organization suggestion.
It can be included in review records according to team-specific procedures and is not a form that replaces approved test plans or product-specific shipping judgments.
